This effect of sennosides (more precisely, their active metabolite product - reynanthron) within the large intestine is determined mainly by the effect on its functional mobility: stationary slows down, and propulsive contractions are simultaneously stimulated. This leads to faster passage of food inside the gastrointestinal tract, which reduces the duration of contact between the chyme and the walls of the intestine, and also reduces the resorption of the fluid.

Along with this, stimulation of the activity of hydrochloric acid release also leads to the secretion of water with electrolytes.

Pharmacokinetics

Glycoside-synthesized cenozides (the so-called caciotides) is a prodrug. They do not lend themselves to splitting and absorption inside the upper part of the digestive tract. These elements are split by the bacterial enzymes inside the large intestine, transforming into a reynanthron.

Reynanthron is a metabolic product that exerts a laxative effect. He has extremely low systemic bioavailability. In animal tests, less than 5% of sennoside and rhein derivatives (in the form of substances subjected to oxidation, and in addition partial conjugation) were excreted together with urine.

A significant part of reynanthron (more than 90%) falls into the feces, and there it is firmly synthesized with intestinal contents. Excretion takes place in the form of compounds of the polymer type.

A small part of the active metabolic products (eg, rhein) passes into the mother's milk. Passage of rhein through the placental barrier during the tests on animals occurred in minimal quantities.

• hepatic lesions of an organic nature;
• disorders of water-electrolyte balance in severe degree (eg, hypokalemia);
• bleeding in the rectal area;
• children under 12 years of age.
